The two-time world champion impressed by cruising through the first round before dominating Japan's Yuta Wakimoto in the best of three quarter-finals.
The 27-year-old is showing no signs of slowing down after backing up from his keirin silver medal won on Saturday night.
He has emerged as an inspiration for the Australian track cycling team after opting to continue racing despite being diagnosed with thyroid cancer in late October.
He will undergo treatment after the Brisbane round which concludes on Sunday night but is hopeful of contesting February's world titles and the Tokyo Olympics.
Glaetzer is aiming to add to Australia's tally of six medals - which includes three gold - on the Brisbane round's final night.
Gold will be on offer in four events on Sunday - women's omnium, men's sprint, men's madison and women's keirin.
Earlier on Sunday, Australia's Stephanie Morton backed up from her sprint silver on Saturday night to cruise into the women's Keirin second round.
She was the sole Australian in the field with 2019 world titles silver medallist Kaarle McCulloch managing a back complaint.
Morton won the 2019 team sprint world title with McCulloch.
Meanwhile, Australia's Nathan Hart failed to qualify for the men's sprint quarter-finals.
The Brisbane round is the Australian track cycling team's final chance to race on home soil before the world titles in Berlin and offers qualifying points for the Tokyo Olympics.
The 18-strong Australian squad features 10 reigning world champions.
