Tall pivot Jono Ford was at his creative best for the Roos while lock Eloni Vunakece hit hard and often in an eye-catching performance.
Ford scored two tries, had a hand in two others, and proved to be a constant threat to the defence.
Vunakece played with the authority and impact befitting a Fijian World Cup player. His stinging defence and incisive charges with the ball added muscle to the Wyong pack.
Wyong led 20-0 at the break and maintained their discipline to keep Manly scoreless in the second half.
Wyong football manager Matt Lavin said it was an impressive win for the home team given the quality of players who sat out the match.
"Jay Florimo, Bennett Leslie and Tekina Vailea were out suspended, skipper Mitch Williams was in camp with the NSW Residents team, and Chris Centrone and Shaune Corrigan were both unavailable due to injury," Lavin said.
The win moves Wyong to 18 points on the NSW Cup ladder, equal with Newcastle and in ninth position.
Wyong has a bye this weekend. They return to Morry Breen Field on Sunday, July 20, and host NZ Warriors.
